An observation of toxic effect of vinblastine on the neurulation stage chick embryo



Abstract
Vinblastine is a well known teratogen as well as an antitumor and antimitotic agent. Present study was undertaken to observe the mechanism by analyzing the correlation of cytotoxicity and embryotoxicity with teratogenic action of this alkaloid.
Chick embryos of white Leghorn were exposed in vivo to hazardous doses of vinblastine sulfate (0.17mg~0.5mg/kg of egg) at the time of early neurulation stage, 18th hour of incubation. Following exposure, the embryos were processed for observation with gross and light microscope.
The extent of the deformities was dose related from growth retardation to dissolution of the embryos.
The cytotoxic effects of vinblastine were manifested in the form of cytoplasmic granulation, swelling of the cells and edematous change of the germ layers. The metaphase arrested cells were accumulated in the lapse of time in all the layers of embryo which became separated each other and detached from their layers and consequently failed to maintain their histoarchiteeture. These observations indicate that vinblastine possibly may affect cell membranes and adhesion secondary to inhibition of specific synthetic activities.
Cells in mesenchymal layer were more severely affected than those in neuroectoderm and those in endoderm and surface ectoderm were far less affected, which suggest that the higher proliferating activity of cell, the more susceptible to the drug.
